The request in this Non-Competitive Action is to upgrade the existing equipment, which entails a hardware change to the camera and compute.

### Procurement Information

The Genetics Core is embedded within the Edinburgh Clinical Research Facility, which has been accredited under the Medicines and Healthcare Products Regulatory Agency (MHRA) Phase I Accreditation Scheme since July 2011. The Genetics Core purchased the original equipment in 2024, which is a relatively new-to-market short-read sequencing platform. The request in this Non-Competitive Action is to upgrade the existing equipment, which entails a hardware change to the camera and compute. This request builds on the initial investment and allows two new functionalities; a) Increased throughput & cost saving: The upgrade enables ~50% increase in sequencing output for no extra reagent or processing cost e.g. High output flow cell increases from 1B reads to 1.5B reads. This results in a cost saving each time the platform is run. The trend of direction in genomics is for higher throughput sequencing, especially in the fields of single-cell (10x, Parse) and spatial biology (Visium, Nanostring GeoMx). b) Simultaneous multi-omic profiling at the single cell level: The upgrade allows the exciting ability to allow in-sample omics profiling so that transcriptomics and proteomics occur within the cell, without the need for DNA or RNA extraction. This in-situ sequencing on the platform allows co-detection of RNA, protein, and cell morphology at subcellular spatial resolution. For example, the curated neuroscience panel available from Element contains RNA, protein & cell painting markers to study neurodegenerative disorders, like Alzheimer's and Parkinson's disease, psychiatric disorders, and neurological cancers. It can unravel the complexities of neuroscience, from understanding inflammation and identifying key drivers of neural cell function to exploring neuronal differentiation and shedding light on disease progression mechanisms. The main new research activity will enable the ability for in-situ multi-omics analysis. This novel technique allows sequencing within living cells without the need for lysis. In addition, the 50% increase in sequencing throughput will allow the Genetics Core to ensure demand is met, the centralisation of genomics and the expected increase in projects and researchers using the Genetics Core. The proposed procurement route is a Non-Competitive Action (NCA), as Element are the only provider that can upgrade their equipment. Further details in this document justify this approach as a compliant route to market.
