Notice Title
F/2627 - FFF/SEC-Multidetection System
Notice Description
The FFF/SEC-multidetection instrument is for separating, characterising, and fractionating both biomacromolecules and biological particles. The biomacromolecules include biopolymers such as polysaccharides (e.g., glycosaminoglycans), proteins, nucleic acids, mucins, while the biological particles include antibodies, nanocomplexes, exosomes, mucus, among other. These systems are to be analysed as pure or as mixtures. The separation is achieved either by size exclusion HPLC (SEC-HPLC) or by asymmetrical flow field-flow fractionation (AsFlFFF) under thermostated conditions. In both cases, this is according to the dimensions given by the molar mass and hydrodynamic size of the polymers and particles. In both separation modes, the system enables the determination of the molar mass, hydrodynamic radius (Rh), radius of gyration (Rg,) polymer conformation and branching degree of biopolymers, as well as the particle size (Rh) of nano- and micro-particles. The characterisation is made by a multidetection system that uses dynamic and multi-angle laser light scattering (DLS and MALS, respectively), while the composition is elucidated by a UV/VIS spectroscopy and a differential refractive index (DRI) detectors. The fractionated eluting polymer and particle species pass through all these detector units arranged online, and the software integrates the information to generate the corresponding chromatograms (SEC) and fractograms (FFF) as well as the molecular mass, Rg, and particle size (Rh) distribution plots and parameters derived thereof. The separated fractions can subsequently be recovered by an automated fraction collector system. Additional information: Is a Recurrent Procurement Type? : No
