Summary of the contracting process

Manchester University NHS Foundation Trust is preparing to procure LAMP (loop-mediated isothermal amplification) assay kits for clinical testing of DPYD, CYP2C19 and HFE variants. The requirement is for chemical reagents and associated assay solutions, ready for clinical use in the Trust’s laboratory operations. Testing must identify the relevant variants listed in the national genomics test directory and distinguish homozygous wild-type, heterozygous mutant and homozygous mutant genotypes. The work will support services delivered in the North West of England, with the buying organisation based in Manchester. Kits must be compatible with the laboratory’s accredited testing environment and provide validated, reliable results for clinical decision-making.

This procurement is at the planning and early market-engagement stage; the buyer has stated an intention to procure, but this is not an invitation to tender. The buyer expects engagement to run until 26 August 2026. The anticipated procurement value is £230,000. The planned purchase is for goods, using an assay-based solution for the three genetic testing requirements. The buyer expects suppliers to provide validation evidence for analytical sensitivity and specificity, with a target of 100% for both, repeatable and reproducible results, and a technical failure rate below 5%. Each sample should be processable end to end within four hours. Suppliers are expected to demonstrate capacity for approximately 6,000 DPYD, 400 CYP2C19 and 7,000 HFE samples each year.

This will suit specialist manufacturers and distributors of molecular diagnostic or genomics assay kits with established clinical laboratory support. Suppliers should be able to provide CE-marked kits suitable for use under UKAS ISO 15189:2022-accredited laboratory arrangements, with the necessary assurance that the solution is ready for clinical deployment. Strong technical evidence will be important, including validation of analytical sensitivity and specificity, detection of nucleotide changes, genotype discrimination, repeatability and reproducibility. The supplier will need operational capacity to support high-volume DPYD and HFE testing alongside lower-volume CYP2C19 testing, while maintaining a technical failure rate below 5% and a four-hour end-to-end sample-processing time. This is an opportunity to prepare capability and evidence before the future tender.

Procurement of LAMP (Loop-mediated isothermal amplification) assay for DPYD, CYP2C19 and HFE testing. Additional information: The bidder must be able to provide an assay which detects the variants for DPYD, CYP2C19 and HFE as stated on the national genomics test directory. Please note: due to the laboratory's UKAS 15189; 2022 accreditation we require any kits to be CE marked. The assay must be ready for clinical use. The bidder must provide full details of the validation of this solution used to determine the values provided for analytical sensitivity and specificity for detection of nucleotide changes included in each assay. Analytical sensitivity and specificity should be 100%. The assay should produce results that are repeatable, with concordant results for samples replicated within runs. The assay should demonstrate reproducibility, where typical test-to-test variability does not impact the result. Concordant results should be achieved for samples replicated across runs. The assay must be validated to detect variants, with a need to differentiate between wild type and mutant alleles, and can therefore detect homozygous wildtype, heterozygous mutant and homozygous mutant genotypes. The technical failure rate should be <5%. The kit should be possible to process a sample within 4 hours, end to end.
