The Centre for Cardiovascular Science (CVS) is part of the new Institute of Neuroscience and Cardiovascular Research (INCR). CVS comprises ~200 researchers encompassing a wide range of cardiovascular research split into four main themes (Metabolism, Obesity and Diabetes; Coronary and Valvular Heart Disease; Hypertension and Renal; Cardiometabolic Imaging). Centre for Cardiovascular Science (CVS) requests an Agilent Seahorse XFe24 analyser and software for high throughput real-time metabolic analysis of living cells and tissue explants. The instrument will add easy-to-use capability for metabolic measurements in living cells to better understand cellular metabolism. The replacement XFe24 machine will support the requirements of the centre to deliver world-leading research in the field of metabolism. The replacement XFe24 will allow high sensitivity measurement of real-time respiration in immortalised and primary cells and in tissue explants in several species.

Procurement Information

Seahorse XF instruments are the only ones that can simultaneously measures oxygen consumption rate (OCR) and extracellular acidification rate (ECAR) of living cells, and do so with unrivalled speed and requiring less material, even than other systems that solely measure OCR. Using these measurements, glycolytic Proton Efflux Rate (PER) is easily calculated as an accurate measurement of glycolysis rates in real time. This measurement technology is covered by US patent # 7,276,351 meaning that no other company can offer this capability. The XFe24 Seahorse is the only commercially available instrument for scientific research that simultaneously measures the two major energy-producing pathways of the cell, mitochondrial respiration and glycolysis. The requested instrument is unique in its offerings and ability to perform measurements in living cells without using destructive methods, thus preserving cellular integrity, and allowing researchers to follow cell fate over time. The XFe24 allows simultaneous analysis of a plate containing 24 wells - and has the ability to study 3D models (e.g. islets, adipose cells and tissue explants) which is crucial for several groups in CVS. Four compounds can be injected sequentially into each of the 24 wells during measurement of cellular respiration (oxygen consumption and extracellular acidification) to determine cellular responses in real time. Unique technical features not found in any other instruments Instrument creates transient microchamber, makes optical measurements, and performs compound injections. Temperature controlled measurement chamber is maintained at 37degC +- 0.5degC; alternative temperatures supported. XFe analyzers are also supported to for analysis in a hypoxic chamber.
